Blood cultures
done when sepsis, endocarditis, meningitis, penuemonia or osteomelitis is suspected
blood cultures organisms most often isolated
gram + (cocci) staph. aureus, strep pneumoniae
gram - (rods) E. coli, K. pneumoniae, P Aeruginosa
what to check for in blood cultures
mixed w/ rich medium & anerobic growth
Growth in blood cultures
gram stain, antibiotic sensitivity
Throat cultures
test for B- hmolytic streptococci
throat culture
test with bacitracin- if growth inhibited prob group A streptococcus---> pyogenes
sputum cultures
performed when pneumonia, TB, or lung abscess suspected
cause of pneumonia in hospital
staph aureus, K. pneumoniae ,or other gram - rods
cause of pneumonia outside hospital
strep. pneumonia gram + diplococcus
Sputum staining
if TB suspected, acid fast stain is done
spinal fluid cultures
performed when meningitis is suspected
acute bacterial meningitis
usually caused by N. meningitis, step pneumoniae or H. influenze
acute staining
gram - stained followed by immunological tests
subacute
usually caused by M. tuberculosis and fugus crytococccus
Subacute staining
spinal fluid acid fast stained and cultured
stool cultures
used in cases of enterocolitis. usually cases of culpritis are shigella, salmonella, and camplyobacter
stool cultures gram staining
usually not useful because of large bacteria flora in colon
Stool staining
methly blue stain if large number of leukocytes present
genital tract cultures- male
presence of garm - diplococci within neutrophils---> N. gonorrhea
genital tract cultures- female
chlamydia trachomatis
syphilis
cultured in human cells or chickin eggs. motile spirochetes
quellung reaction
swells when specific antibody binds
slide agglutination test
specific antibodies against cell wall are mixed with cells on microscope slide. form large clumps
macConkey Agar or EMB agar
Inhibit gram +
urine cultures
most common case is E. coli, others are enterobacter, proteus, strep faecalis
asynptomatic patients
at least 100,000 cells/mL
symptomatic patients
need at least 100 cells/mL
ELISA
anitgens detected w/ specific antibody that has enzyme attached at Fc end
Serological techniques
isolate blood serum containing antibodies of unknown specifity and react with antibodies against known organisms
PCR
use ssDNA primers to amplify gene from organism's genome
Hybridization
hydridize ssDNA with probe attached to organism's DNA
Nucleic acid sequencing
determinine sequence of DNA or RNA from organism
