Recombinant DNA
Nucleotide sequences from two different sources, often two species, are combined into the same DNA molecule.
DNA Cloning
*general method involves 2 main tools: bacteria and their plasmids.
Genome fragments are copied into a easily manipulated form.
Plasmid
small circular DNA molecule that replicate separately from bacterial chromosome.
Restriction enzyme
Bacterial enzymes that cut DNA molecules at specific DNA sequences, restriction sites.
Restriction site
A specific sequence on DNA strand that is recognized and cut by a restriction enzyme.
DNA ligase
covalently joins the fragment and plasmid.
Bacterial Transformation
The conversion of a normal animal cell to a cancerous cell.
Genomic Libraries
thousands of individual bacteria clones, each with a plasmid holding a unique DNA fragment from an organism's genome.
Complimentary DNA (cDNA)
A double stranded DNA molecule made in vitro using mRNA as a template and the enzymes reverse transcriptase and DNA polymerase.

Nucleic acid hybridization
method of identifying a specific DNA sequence.
Polymerase chain reaction (PCR)
produces many copies of a specific segment of DNA
Dideoxynucleotide
is a DNA nucleotide that has an oxygen atom removed from its 3' carbon.

a collection of scientific methods for determining the sequence of the nucleotide bases in a molecule of DNA.
