M1 Muscarinic Receptors
LOCATION: CNS neurons, ganglion cells near parietal cells of stomach
SIGNALING: phospholipase C --> increase IP3 --> increase intracellular calcium
SIGNIFICANCE: Alzheimer's disease, ulcers
M2 Muscarinic Receptors
LOCATION: heart, postganglionic autonomic nerve endings, smooth muscle
SIGNALING: activation of K+ channels --> inhibition of adenlyl cyclase
SIGNIFICANCE: cardiac slowing, modulation of transmitter release
M3 Muscarinic Receptors
LOCATION: exocrine glands, smooth muscle, endothelium
SIGNALING: phospholipase C --> increase IP3 --> increase intracellular calcium
SIGNIFICANCE secretion, smooth muscle contraction, vasodilation at small doses
Methacholine
Muscarinic AGONIST
Use: diagnostic, assess bronchiolar reactivity
